Abstract

DNA methylation plays a critical role in the epigenetic regulation of gene expression, and 5-methylcytosine (5mC) is one of the most important biomarkers of this process. Rapid and reliable detection of 5mC in the presence of cytosine is of great importance in biomedical research and clinical diagnosis. In this study, an electrochemical method was developed for the determination of 5mC. Different voltammetric techniques, especially differential pulse voltammetry (DPV) and square wave voltammetry (SWV) were used to investigate the electrochemical behaviour of 5mC. The study investigated the redox properties of 5mC in different pH media, in the range of 2.00–11.00 on disposable PG electrodes. The results show that the proposed method for voltammetric determination of 5mC in the presence of cytosine is an environmentally friendly and practical tool for monitoring epigenetic changes. This study offers a cheaper and eco-friendly approach for epigenetic research and clinical applications, showing potential in the diagnosis and treatment monitoring of diseases related to DNA methylation.
