Samples were processed with a Density Gradient Centrifuging. Spermatozoa were divided into 4 groups: control, experimental 1, 2 and 3 (E1, E2, E3) based on the dose/time schedules (cAMP 5mmol<!--> <!-->+<!--> <span>IBMX 0.2mmol/2, 4, and 6</span> <!-->h, respectively). The computer-assisted sperm analysis and chlortetracycline assays were used to measure sperm motility and capacitation.</p></div><div><h3>Results</h3><p>After incubation with a cAMP analog and IBMX, the levels of progressive motile sperms considerably improved in all experimental groups compared to the control group (E1<!--> <!-->=<!--> <!-->18.89<!--> <!-->±<!--> <!-->7.1, E2<!--> <!-->=<!--> <!-->30<!--> <!-->±<!--> <!-->9.7, E3<!--> <!-->=<!--> <!-->26.3<!--> <!-->±<!--> <!-->9.6 vs Control<!--> <!-->=<!--> <!-->10.28<!--> <!-->±<!--> <!-->6.2, <em>P</em> <!-->&lt;<!--> <!-->0.05) especially in E2 group (<em>P</em> <!-->&lt;<!--> <!-->0.05), indicating a greater effect of db cAMP (5<!--> <!-->mmol) and IBMX (0.2<!--> <!-->mmol) for 4<!--> <!-->h compared to the same doses at 2 and 6<!--> <!-->h. 摘要

目的:人类精子质量日益下降。不孕不育的主要原因之一是精子失能。我们研究了cAMP(环腺苷单磷酸类似物)+ IBMX(3-异丁基-1-甲基黄嘌呤)随时间对人类精子活力和获能率的影响。材料和方法样本取自伊朗库姆不孕症研究中心教育、文化和研究学术中心的20例弱精子不孕症患者。样品用密度梯度离心处理。按剂量/时间(cAMP 5mmol + IBMX 0.2mmol/2、4、6 h)将精子分为对照组、实验1、实验2、实验3 (E1、E2、E3) 4组。使用计算机辅助精子分析和金四环素测定精子活力和获能性。结果经cAMP类似物和IBMX孵育后,各试验组的进行性运动精子水平均较对照组显著提高(E1 = 18.89±7.1,E2 = 30±9.7,E3 = 26.3±9.6 vs对照组= 10.28±6.2,P <0.05),尤其是E2组(P <0.05),表明db cAMP (5 mmol)和IBMX (0.2 mmol)作用4 h比相同剂量作用2和6 h的效果更大。此外,E2组的非进行性运动精子显著减少(P <0.05)。此外,所有实验组的C型和B型均有明显改善,尤其是E2组(P <0.05)。结论db cAMP + IBMX对弱精子症患者的辅助生殖成功率有一定的促进作用,特别是对弱精子症患者补充4 h。

Cyclic adenosine monophosphate (cAMP) analog and phosphodiesterase inhibitor (IBMX) ameliorate human sperm capacitation and motility

Objectives

Human sperm quality is decreasing progressively. One of the foremost reasons for infertility is the failure in sperm capacitation. We examined the influence of a cAMP (cyclic-adenosine mono phosphate analog) + IBMX (3-isobutyl-1-methylxanthine) on the motility and capacitation rate of human sperm over time.

Material and methods

Samples were gotten from 20 asthenozoospermic infertile patients referring to the Academic Center for Education, Culture and Research unit of the infertility research center, Qom, Iran. Samples were processed with a Density Gradient Centrifuging. Spermatozoa were divided into 4 groups: control, experimental 1, 2 and 3 (E1, E2, E3) based on the dose/time schedules (cAMP 5mmol + IBMX 0.2mmol/2, 4, and 6 h, respectively). The computer-assisted sperm analysis and chlortetracycline assays were used to measure sperm motility and capacitation.

Results

Conclusion
