摘要

单克隆抗体彻底改变了癌症和自身免疫疾病患者的治疗方式,是当代药物开发的最前沿。然而,由于抗体体积较大,无法有效渗透到细胞质中,因此传统上认为抗体无法有效靶向细胞内抗原。然而,人们很早就知道,IgG 可通过与 FcRn 结合和内泌体的胞内转运穿过胎盘,这种现象被称为转胞吐。此外,二聚体 IgA 和五聚体 IgM 也以聚合免疫球蛋白受体(PIGR)依赖的方式通过粘膜细胞进行定向转运。这一复杂的过程不仅能将这些免疫球蛋白转运到粘膜位置的管腔侧,还能中和已经感染上皮细胞的病毒[1]。
