At the same time thrombin induces the profound changes in the levels of platelet glycolytic intermediates with the \"crossover\" point at phosphofructokinase step of glycolysis. 摘要

本实验旨在研究凝血酶对洗涤后人血小板糖酵解的作用机制。从人血中分离的血小板,用Krebs-Ringer-phosphate缓冲液(pH 7.4)重悬,在水浴(37℃)中孵育,不加(对照)或加凝血酶(1 NIH U/ml血小板悬液)。采用酶分析和荧光法测定血小板中和的高氯酸提取物中的乳酸、糖酵解中间体和腺嘌呤核苷酸。本研究结果表明,凝血酶可显著提高洗涤后的人血小板乳酸生成率。同时凝血酶以糖酵解磷酸果糖激酶步骤的“交叉点”诱导血小板糖酵解中间体水平发生深刻变化。结果表明,凝血酶刺激血小板糖酵解通过变构磷酸果糖激酶激活,由于腺嘌呤核苷酸的能量电荷的变化。
