The results showed that ribozyme significantly suppressed the clonogenic growth and the expression of p210 protein in primary cells from CML patients, but did not affect the growth of normal hematopoietic progenitor cells. In the model of remission, ribozyme eliminated the proliferation and the expression of bcr-abl mRNA in residual K562 cells, but did not affect the expression of abl mRNA. 摘要

为了研究锤头核酶对慢性髓性白血病(CML)细胞和骨髓清除的影响,我们通过逆转录病毒转导将bcr-abl特异性核酶基因导入CML和正常骨髓细胞。采用造血祖细胞法、流式细胞术和免疫细胞化学方法检测核酶对CML患者原代细胞的影响。建立缓解模型,用核酶转导模型后,采用白血病集落法和巢式pcr检测核酶对骨髓清除的影响。结果表明,核酶能显著抑制CML患者原代细胞的克隆生长和p210蛋白的表达,但不影响正常造血祖细胞的生长。在缓解模型中,核酶消除了残余K562细胞的增殖和bcr-abl mRNA的表达,但不影响abl mRNA的表达。提示抗bcr-abl核酶可用于CML细胞的骨髓纯化。
