Myocardial ischemia-reperfusion was created by ligating the left anterior descending coronary artery for 30 min followed by 3 h of reperfusion. Myocardial infarction sizes were determined by dual staining with triphenyltetrazolium chloride and trypan blue. Plasma levels of MMP-2 were measured using ELISA. Myocardial MMP-2 messenger RNA was analyzed by reverse transcription polymerase chain reaction.</p><p><strong>Results: </strong>The mean (± SD) infarct size in the ischemic postconditioning group was significantly smaller compared with the ischemia-reperfusion group (37.1±3.8% versus 57.5±1.9%; P=0.02). The incidence of ventricular tachycardia in the ischemic postconditioning group was also lower than in the ischemia-reperfusion group (8.5% versus 75%; P=0.003). 目的:探讨缺血后处理对兔心肌缺血再灌注过程中基质金属蛋白酶(MMP)-2表达的影响。方法:36只雄性新西兰大白兔随机分为假手术组、缺血再灌注组和缺血后适应组。结扎左冠状动脉前降支30min,再灌注3h,形成心肌缺血再灌注。心肌梗死大小采用三苯基四唑氯和台盼蓝双染色法测定。ELISA法检测血浆MMP-2水平。逆转录聚合酶链反应分析心肌MMP-2信使RNA。结果:缺血后适应组的平均梗死面积(±SD)明显小于缺血再灌注组(37.1±3.8% vs 57.5±1.9%;P = 0.02)。缺血后适应组室性心动过速的发生率也低于缺血再灌注组(8.5%对75%;P = 0.003)。缺血后适应组MMP-2信使RNA表达明显低于缺血再灌注组(0.4944±0.0476 vs 0.6989±0.0694;P = 0.02)。结论:缺血后处理可能通过抑制MMP-2的表达而减轻心肌缺血再灌注损伤。
