Historically, KRAS mutations have been difficult to target due to lack of binding sites and exceptionally high affinity for guanosine triphosphate/guanosine diphosphate (GTP/GDP). Recently, KRAS<sup>G12C</sup> selective inhibitors have shown promising results in Phase I/II studies.
