摘要

目的探讨烟酰胺腺嘌呤二核苷酸磷酸(NADPH)对大鼠心肌缺血再灌注(I/R)损伤的影响。方法选用SPF级成年雄性sd - dawley大鼠56只,体重220 ~ 320 g, 1 ~ 2月龄,采用随机数字表法分为4组(n=14):假手术组(sham组)、心肌I/R组(I/R组)、NADPH组(n组)和地尔硫平组(D组)。麻醉大鼠左前降支结扎30min,再灌注2h,建立心肌I/R损伤模型。N组从再灌注5 min开始,5 min内静脉滴注NADPH 16 mg/kg。地尔硫卓5 mg/kg于缺血前10 min经颈内静脉滴注至缺血结束。再灌注2 h取颈内静脉血,测定血清LDH和cTnI浓度,取心肌组织测定梗死面积和ROS水平。结果与Sham组比较,I/R组大鼠血清LDH、cTnI浓度、心肌梗死面积、ROS水平均显著升高(P < 0.05)。结论NADPH可通过抗氧化作用减轻大鼠心肌I/R损伤。关键词:NADPH;心肌再灌注损伤
