摘要

目的:蛋白质组学差异的测定在生物标志物研究中起着重要作用。由于其分子背景的异质性,识别某些生物标志物仍然是神经母细胞瘤诊断和预后的需要。在本研究中,旨在确定一些可能在神经母细胞瘤预后中发挥作用的标志蛋白/机制。方法:采用基质辅助激光解吸电离-飞行时间(MALDI-TOF/TOF)方法对不同的疾病风险组进行蛋白质组学研究。构建了来自风险组患者血液样本的单核细胞池,并鉴定了不同组的蛋白质表达变化。对这些患者的N-MYC、11q、1p和17q状态进行实时PCR分析,并从肿瘤样本中确定风险组。结果:神经母细胞瘤高危组患者血清锰超氧化物歧化酶(SOD2)蛋白明显升高。结论:SOD2可能在神经母细胞瘤的进展中发挥重要作用,是神经母细胞癌患者预后的候选外周血标志物。
