摘要

目的研究长链非编码RNA HIF1A-AS1对心肌缺血再灌注(I/R)损伤的调控作用及其机制。方法用SD大鼠建立心肌I/R损伤模型,用大鼠心肌细胞建立缺氧再氧化(H/R)模型。使用si-HIF1A-AS1抑制HIF1A-AS1在心肌中的表达。实时荧光定量PCR检测HIF1A-AS1 mRNA表达,MTT法检测心肌细胞生长活力,ELISA法检测培养基中乳酸脱氢酶(LDH)浓度,Western blotting法检测自噬相关蛋白beclin -1表达。结果HIF1A-AS1在I/R模型大鼠心肌和H/R模型大鼠心肌细胞中表达升高。siRNA抑制HIF1A-AS1可保护心肌细胞免受H/R损伤,逆转H/R刺激诱导的心肌生长活力下降、培养基中LDH水平升高、自噬相关蛋白beclin -1表达增加。结论抑制长链非编码RNA HIF1A-AS1可能通过抑制心肌细胞过度自噬对心肌I/R损伤起保护作用。
