摘要
目的观察β-arrestin2在卵巢切除(OVX)大鼠心脏组织中的表达,探讨β-arrestin2在雌激素介导的OVX大鼠心血管保护中的作用。方法通过监测OVX大鼠血清雌激素水平和子宫重量与体重之比对其进行评价。雌激素替代治疗4周后,通过测量大鼠的心重与体重之比、血压和心率来评估大鼠的心血管功能,采用Western blotting分析检测心脏组织中β-arrestin1、β-arrestin2和血管紧张素受体1型(angiotensin receptor type 1, AT1R)蛋白的表达。结果与假手术组比较,OVX大鼠血清雌激素水平、子宫重量/体重比均显著降低(P0.05); OVX大鼠血压、心率、心重/体重比均显著升高(P0.05),经雌激素替代治疗后均有所降低。与假对照组相比,OVX大鼠心脏β-arrestin2表达显著降低(P0.05),雌激素表达上调,β-arrestin1和AT1R表达不变。结论OVX大鼠心脏组织中β-Arrestin2表达降低,雌激素替代治疗可使其表达升高。
