The establishment of a suitable in situ model of glioma is the premise of the treatment of glioma. 8 week-old female BALB/c nude mice were chose to establish the glioma model. Methods: For the orthotopic glioma mice model, 1 × 105cells/5 μL U87-MG-Luc or U87-MG cells which were trypsinized and resuspended in sterile PBS were slowly injected into the right corpus striatum (1.8 mm lateral, 0.6 mm anterior to the bregma and 3.0 mm in depth) by a stereotactic fixation device using a mouse adaptor. Results: The othotopic U87 glioma mice model identified by imaging on IVIS Spectrum and magnetic resonance imaging after 2 weeks from surgery. H&E-stained tumor sections in brain of the mice model were also observed. 摘要

背景:由于胶质瘤的特殊位置,手术和化疗是困难的。建立合适的胶质瘤原位模型是胶质瘤治疗的前提。选择8周龄雌性BALB/c裸鼠建立胶质瘤模型。方法:将1 × 105个细胞/5 μL U87-MG- luc或U87-MG细胞经胰蛋白酶化、重悬于无菌PBS中,用小鼠接头用立体定向固定装置缓慢注射于右侧纹状体(外侧1.8 mm、内侧0.6 mm、内侧3.0 mm)。结果:术后2周,采用IVIS谱和磁共振成像技术建立了异位U87胶质瘤小鼠模型。h&e染色小鼠模型脑内肿瘤切片。结论:经鉴定获得的胶质瘤小鼠异种移植原位模型可用于胶质瘤治疗药物或方法的评价体系。
