摘要

目的:选择一种合适的聚合物,用包埋法固定化胰猪脂肪酶,并用游离酶评价固定化酶的动力学性质。方法:。采用海藻酸钙和海藻酸钡聚合物包埋法固定化酶。用滴定法测定脂肪酶活性。Km和Vmax用Michaelis Menton方程确定。结果:与藻酸钡微球相比,负载酶的藻酸钙微球在高温下更稳定,具有优良的Vmax和Km。与固定化酶相比,游离酶不耐高温。结论:海藻酸钠聚合物是胰猪脂肪酶包埋的优良载体。
