Methods: H9c2 myocardial cells were randomly divided into four groups: control group, hypoxia reoxygenation model group (H/R), Exenatide group and PI3K/AKT signaling pathway inhibitor group. CCK-8 method was used to detect myocardial enzymes, and the expression of Caspase-3, Bcl-2 and Bax proteins was detected by Western blot. Results: The apoptosis rate in exenatide group was decreased than the H/R group. The levels of LDH and CK-MB in H/R group were higher than control group, which compared with the H/R group, exenatide group were decreased, and it in PI3K/AKT pathway inhibitor group were increased than exenatide group. The expression of Caspase-3 and Bax in H/R group was higher than that in control group group, and the level of Bcl-2 was decreased. Compared with H/R group, the levels of Caspase-3 and Bax in exenatide group were decreased, and the level of Bcl-2 was increased. Compared with exenatide group, the levels of Caspase-3 and Bax were significantly increased in the PI3K/AKT pathway inhibitor group, and the level of Bcl-2 was decreased. Conclusion: Exenatide has protective effects on the cardiomyocytes during hypoxia reoxygenation injury. 摘要

目的:观察GLP-1R激动剂对缺氧复氧心肌细胞的保护作用,并探讨GLP-1R/PI3K/AKT信号通路的作用。方法:将H9c2心肌细胞随机分为4组:对照组、缺氧复氧模型组(H/R)、艾塞那肽组和PI3K/AKT信号通路抑制剂组。采用CCK-8法检测心肌酶,Western blot检测Caspase-3、Bcl-2、Bax蛋白的表达。结果:艾塞那肽组细胞凋亡率明显低于H/R组。H/R组LDH、CK-MB水平高于对照组,与H/R组比较,艾塞那肽组降低,PI3K/AKT通路抑制剂组较艾塞那肽组升高。H/R组Caspase-3、Bax表达高于对照组,Bcl-2表达降低。与H/R组比较,艾塞那肽组Caspase-3、Bax水平降低,Bcl-2水平升高。与艾塞那肽组比较,PI3K/AKT通路抑制剂组Caspase-3、Bax水平显著升高,Bcl-2水平降低。结论:艾塞那肽对心肌细胞缺氧再氧化损伤有保护作用。GLP-1R/PI3K/AKT信号通路可能参与了艾塞那肽抑制心肌细胞凋亡的过程。
