The role of CD44 in tumorigenesis is due to its binding to extracellular matrix components, including hyaluronan (HA) and osteopontin (OPN) and to messenger molecules, such as growth factors present in the tumor microenvironment where they contribute to the maintenance of the stemness of malignant cells. Nanog and Oct are pluripotent stem cells that play roles in development of cancer stem cell. The aim of this research was to investigate expression of CD44, Nanog and Oct 4 at colorectal patient stadium IV. Biopsy of patient was extracted using collagenese I and incubate at 37°C for 30 minutes. Upon biopsy, the tissues were analyzed using Tali cytometer and indirect immunostaining for CD44, Nanog and OCT 4 expressions at days 3, 7 and 10. The results were compared with that of colo cell line. Results revealed the significance differences in gene expression between samples from patients with line colo cells originating from Caucasian colon cancer patients.
