Methods : Immunization of mice with a synthetic peptide derived from the IL-23p19 sequence and generation of hybridoma secreting specific antibodies. The chimeric antibody was created using two eukaryotic plasmid constructions; one of them carrying the light mouse-human chain and the other the heavy mouse-human chain. CHO-K1 cells were cotransfected with both plasmids and stable transfectants were grown in selective culture medium. Results : A chimeric version of anti-IL-23p19 was successfully constructed and expressed in eukaryotic cells. The expressed chimeric antibody showed specific recognition not only of the peptide used as immunogen but also the subunit p19 and the complete interleukin Il-23. 摘要

目的:利用合成的肽段作为免疫原,构建表达人-鼠嵌合抗IL-23p19抗体。方法:用IL-23p19合成肽段免疫小鼠,产生分泌特异性抗体的杂交瘤。利用两个真核质粒构建嵌合抗体;其中一个带着轻的鼠人链,另一个带着重的鼠人链。用两种质粒共转染CHO-K1细胞,并在选择性培养基中培养稳定的转染物。结果:成功构建抗il -23p19嵌合蛋白并在真核细胞中表达。所表达的嵌合抗体不仅能特异性识别作为免疫原的肽,还能识别亚基p19和完整的白细胞介素Il-23。结论:合成的嵌合抗体能够识别具有生物活性的亲本蛋白il - 23,可用于慢性炎症疾病的靶向治疗。
