摘要

以5年生油松×油松当年嫩茎为外植体,采用正交试验对油松×油松的离体生长条件进行优化,成功获得了油松幼苗。结果表明:(1)在添加2.0 mg/L n6 -苄ladenine (6-BA)、0.10 mg/L α -萘乙酸(NAA)和30 g/L蔗糖的DCR培养基中,茎上至茎下1.5~3.0 cm处的片段早期分化率最高,为43.7%;(2)在试验条件中,添加2.5 mg/L 6-BA和0.2 mg/L NAA的DCR培养基最适合不定芽的繁殖,其繁殖率和倍数分别为96.7%和6.2;(3)添加0.3 mg/L NAA、1.5 g/L活性炭和20 g/L蔗糖的DCR培养基最适合不定芽的伸长,其繁殖率为444.6%;(4)将幼苗移栽到添加2.0 mg/L NAA的1/2 DCR培养基上,黑暗培养10 d,然后以40 μmol·m-2·s-1光照12 h/d,生根率高达55.3%。
