The cell proliferation index decreased and apoptosis rate increased, which were significantly different compared with the control ones (P<0.01); (3) The typical DNA Ladder-shaped of cerebrum neurocytes of chickens in experimental groups was displayed by agarose gel electropho- resis. 摘要

采用固定体积、不同梯度三氯化铝(AlCl3)连续腹腔注射的方法,建立了鸡不同程度亚慢性铝中毒模型。采用琼脂糖凝胶电泳、AO/EB双荧光染色和流式细胞术检测大脑神经细胞凋亡情况。结果表明:(1)荧光显微镜下观察到典型的脑神经细胞凋亡和坏死,且各实验组的凋亡指数显著高于对照组(P<0.01),且随铝浓度的升高而升高;(2)大脑神经细胞数量在G0/G1期增加,在G2/M期和S期减少。细胞增殖指数降低,细胞凋亡率升高,与对照组相比差异极显著(P<0.01);(3)琼脂糖凝胶电泳显示了实验组鸡大脑神经细胞典型的DNA阶梯状结构。结果表明,铝能引起大脑细胞DNA损伤,抑制细胞增殖,诱导细胞凋亡。
