Methods: Pulmonary fibrosis model in mice was established by bleomycin (BLM) induction. This study was divided into 7 groups, bone marrow mesenchymal stem cells (BMSCs) as the treatment measure in 4 groups, the saline and pirfenidone as other 2 groups. The body weight of mice after BLM modeling was measured. The content of hydroxyproline (HYP) and collagen 1 (COL1) in lung tissue were determined by kits. Pathological changes of lung tissue were observed by hematoxylin-eosin (HE) staining. The levels of cytokines in serum and lung tissue of mice were detected by Enzyme-Linked Immunosorbent Assay (ELISA) kits. Immunohistochemistry was used to detect the expression of collagen-1 and α-SMA protein in lung tissue of mice. The levels of TGF-β/smad-3 and NLRP3/NF-κB signal pathway in lung was detected by western blotting. Results: BMSCs significantly decreased the content of HYP and COL1 in lung tissue of mice. BMSCs significantly decreased cytokines in serum and lung tissue. Immunohistochemistry results shown BMSCs significantly decreased the levels of collagen-1 and α-SMA in lung tissue. In addition, BMSCs significantly inhibited TGF-β/smad-3 and NLRP3/NF-κB signal pathway in lung tissue. 摘要

目的:探讨骨髓间充质干细胞(BMSCs)对博莱霉素诱导小鼠肺纤维化的影响。方法:采用博来霉素(BLM)诱导小鼠肺纤维化模型。本研究分为7组,以骨髓间充质干细胞(BMSCs)为治疗手段4组,生理盐水和吡非尼酮为治疗手段2组。测定BLM造模后小鼠的体重。采用试剂盒法测定肺组织中羟脯氨酸(HYP)和胶原蛋白1 (COL1)的含量。苏木精-伊红(HE)染色观察肺组织病理变化。采用酶联免疫吸附试验(ELISA)试剂盒检测小鼠血清和肺组织中细胞因子水平。采用免疫组化方法检测小鼠肺组织中胶原-1和α-SMA蛋白的表达。western blotting检测肺组织TGF-β/smad-3及NLRP3/NF-κB信号通路表达水平。结果:骨髓间充质干细胞显著降低小鼠肺组织中HYP和COL1的含量。骨髓间充质干细胞显著降低血清和肺组织细胞因子。免疫组化结果显示,骨髓间充质干细胞显著降低肺组织中胶原-1和α-SMA的水平。此外,骨髓间充质干细胞显著抑制肺组织TGF-β/smad-3和NLRP3/NF-κB信号通路。结论:骨髓间充质干细胞能有效抑制博莱霉素诱导的小鼠肺纤维化,其机制可能与抑制TGF-β/smad-3和NLRP3/NF-κB信号通路的激活有关。
