裂口胸高迁移率组1的表达谱及炎症激活分析
目的:高迁移率组框1 (HMGB1)是一种高度保守的核蛋白,参与硬骨鱼对病原体的免疫反应。本研究研究了鲤科鱼类裂腹鱼(Schizothorax prenanti, SpHMGB1)中HMGB1的结构和功能。方法:采用cph1模型预测SpHMGB1的空间结构。采用定量反转录PCR检测SpHMGB1在不同组织及无乳链球菌感染中的mRNA表达。用合成的SpHMGB1-B框肽处理巨噬细胞,分析其炎症活性。结果:在结构上,SpHMGB1与斑马鱼和小鼠相比具有保守的A盒、B盒和酸尾。SpHMGB1在各组织中普遍表达,其中中肾表达量最高。在体内,SpHMGB1在血液和脾脏中对无乳链球菌感染有明显的诱导反应。合成SpHMGB1-B盒肽激活呼吸爆发,上调巨噬细胞中白细胞介素-1β、肿瘤坏死因子α、白细胞介素-10、干扰素调节因子1、干扰素调节因子7、C-X-C基序趋化因子配体11-1、C-X-C基序趋化因子配体11-2、toll样受体4的信使RNA表达。结论:本研究提示SpHMGB1参与了对细菌病原菌的应答,SpHMGB1- b盒肽在介导葡萄球菌免疫应答中发挥了重要作用。
