日本鱼氏爱德华菌与鳗氏爱德华菌的表型差异
目的:迟发爱德华氏菌是日本养殖海鱼和淡水鱼中引起爱德华氏菌病的病原体。本研究将患病橄榄比目鱼(parichthys olivaceus)分离得到的一株类迟缓棘球绦虫(E. tarda)归为鱼鲆(E. piscicida),将患病红鲷(Red Seabream Pagrus major)分离得到的一株类鳗绦虫(E. anguillarum)归为鳗鲡(E. anguillarum)。本研究旨在了解淡鱼和鳗的表型差异。方法:以14株鱼伊蚊和7株鳗伊蚊为研究对象。每个分离的菌落在脑-心灌注琼脂板上生长,然后进行DNA提取。提取的DNA用PCR扩增。采用API 50 CH检测试剂盒检测分离菌的碳水化合物发酵。此外,在确定的培养基中检查了这两个物种的生长情况。采用高效液相色谱-质谱联用技术对橄榄比目鱼和红鲷血清中的游离氨基酸进行检测和定量。使用Welch's t检验分析游离氨基酸浓度的统计差异。结果:API 50ch检测结果显示,鳗鲡分离菌可发酵l -阿拉伯糖和d -甘露醇,而鱼腥鱼分离菌不发酵。此外,在不添加蛋氨酸和硫酸铁的培养基中,piscicida和eanguillarum的生长受到抑制。在不含苯丙氨酸、酪氨酸、丙氨酸或烟酸的培养基中,piscicida的生长受到抑制,而在不含丝氨酸、半胱氨酸、亮氨酸、苏氨酸或异亮氨酸的培养基中,鳗鲡的生长受到抑制。橄榄比目鱼的血清中酪氨酸和丙氨酸含量较高,而红鲷的血清中苏氨酸和异亮氨酸含量较高,表明鱼鲷和鳗鲷的生长条件较好。结论:本研究确定了一种可用于开发抗鱼母绦虫和鳗母绦虫疫苗的最小定义培养基。
