从海洋珊瑚中分离的类固醇衍生物抗鲍曼不动杆菌的作用。
导言:海洋生物占全球总多样性的一半以上。其中,软珊瑚是广泛分布于珊瑚礁中的重要海洋无脊椎动物群。Sacophyton属是一种重要的植物属,含有具有多种生物活性的大环二萜(樟脑尤其是去樟脑衍生物)、倍半萜和三萜(甾体)等有价值的化合物。一些研究表明,大环二萜和类固醇具有抗炎、抗肿瘤、抗肿瘤和抗菌作用。方法与材料:采用浸渍法,先用丙酮干燥,再用甲醇提取。提取液用正己烷、甲醇、乙酸乙酯、丁醇和水溶剂进行分离。用二氯甲烷:乙酸乙酯(从100:0到80:20)和己烷:乙酸乙酯(从97:3到55:45)的闪速柱色谱法分别对己烷部分和甲醇部分进行分析。经过几个步骤,化合物1、2和3被分离出来并通过1H-NMR进行了表征。在鉴定这些化合物后,分别用连续稀释法、羰基氰化物氯苯肼法和琼脂乙基溴侧轮法测定了分离类固醇的最小抑制浓度和外排泵活性。结果:通过1H-NMR和GC-MS的光谱分析,鉴定出3种甾体。鉴定出化合物1:麦角甾醇,化合物2:麦角-5-en-3β-醇,化合物3,5,8 -外延-5α,8α-麦角- 6,22e -二烯-3β-醇。这些化合物对鲍曼不动杆菌的检测结果表明,这些化合物通过外排泵抑制对鲍曼不动杆菌有显著的作用。结论与讨论:鲍曼不动杆菌是一种广谱耐药细菌。抑制外排泵是对抗细菌耐药性的关键机制。已知鲍曼不动杆菌可引起院内感染,特别是在重症监护病房。戈尔戈斯坦和麦角甾醇衍生物的外排泵抑制作用已被证实。石珊瑚属,尤其是S. tenuispiculatum,是一种发现于波斯湾的珍贵珊瑚。由于海洋珊瑚的药用特性尚未被充分了解,它们可能是解决抗生素耐药性和医院感染的有希望的抗菌剂新来源。
