The medium was composed of three basal media, i. e. Dulbecco's modified eagle (DME), F-12 (Ham) and RPMI-1640, and it was enriched with other supplements. The medium was applied with 1% gelatin and 3% AB serum for CRTC (concentrate rotary tissue culture) system, or with 1% AB serum for hollow-fiber system in LAK cells culture. It was demonstrated that the medium can be used to proliferate LAK cells to about thirty-fold the initial cell number with CRTC system, or to twenty one-fold with hollow-fiber system. 低血清培养基在淋巴因子活化杀伤细胞(lak)细胞密集培养中的应用
中村佳彦、若松中美、小林信正、Watanabe胜人输血中心Noto隆、德田丰、田岛智夫、三美敏夫采用新研制的低血清培养基培养淋巴因子活化杀伤细胞(LAK)。培养基由Dulbecco's modified eagle (DME)、F-12 (Ham)和rpm -1640三种基础培养基组成,并添加其他培养基进行富集。在CRTC(浓缩旋转组织培养)体系中加入1%明胶和3% AB血清,在LAK细胞培养中加入1% AB血清。结果表明,该培养基可使LAK细胞在CRTC体系中增殖到初始细胞数的30倍左右,在空心纤维体系中增殖到初始细胞数的21倍左右。因此,该培养基可用于这些培养体系,在低血清浓度下获得大量LAK细胞。
