Abstract
The role of the liver in the alteration of drugs and other exogenous chemical agents and its relation to the toxic effects of these substances have been reviewed.
In patients with hepatic disease some therapeutic agents may produce an adverse effect by exaggerating the usual physiologic one. There are several mechanisms by which hepatic disease may be produced by exogenous chemical agents. Some substances act as protoplasmic poisons, with a particular tendency to produce hepatic necrosis and parenchymal failure. Others damage the liver by acting as metabolic antagonists or inhibitors of specific hepatic functions. The jaundice produced by some drugs appears to be a manifestation of hypersensitivity.
The syndromes of hepatic disease have been classified as hepatocellular, pure and mixed; hepatocanalicular, pure and mixed, and canalicular. An awareness of the type of syndrome produced is helpful in making the diagnosis, gauging the prognosis and selecting the treatment.

Designed for primary care physicians, each issue of Disease-a-Month presents an in-depth review of a single topic. In this way, the publication can cover all aspects of the topic - pathophysiology, clinical features of the disease or condition, diagnostic techniques, therapeutic approaches, and prognosis.
