Luteineizing hormone (LH) receptor, estradiol level and the number of de Graaf follicles measurement were done by immunohistochemical test, ELISA method, and hematoxilin eosin staining, respectively. Results: The results showed that there was no significant difference found on Mab-hZP3 interaction between 20 μg-60 μg dosages and 10–20 days of observation time towards LH receptor expression, estradiol level and and the number of de Graaf follicles. This was related to specificity of monoclonal antibody used. Conclusion: Mab-hZP3 did not lower LH receptor expression, estradiol level and the number of de Graaf follicles quantity, which means that Mab-hZP3 did not interfere at folliculogenesis and not altering hormones profile. Abstract

目的:研究抗人透明带3单克隆抗体(Mab-hZP3)对小鼠卵巢促黄体生成素(LH)受体表达、雌二醇水平及卵泡数量的影响。材料与方法:采用真实实验后测纯对照组设计作为研究方法。采用对照(Tris HCl佐剂)和单抗- hzp3,剂量分别为20、40、60 μg,分别于第10、15、20天检测。分别采用免疫组化法、ELISA法和苏木素伊红染色法测定黄体生成素(LH)受体、雌二醇水平和de Graaf卵泡数量。结果:结果显示,20 μg-60 μg剂量的Mab-hZP3相互作用对黄体黄酮受体表达、雌二醇水平和de Graaf卵泡数量的影响与观察时间10-20 d无显著差异。这与单克隆抗体的特异性有关。结论:Mab-hZP3不降低黄体生成素受体表达、雌二醇水平和de Graaf卵泡数量,说明Mab-hZP3不干扰卵泡发生,不改变激素谱。由此可见,Mab-hZP3具有作为一种安全的免疫避孕材料的潜力。
