Hepatic stellate cells (HSCs) are vital and have a significant importance in their emerging role in hepatic regeneration.Aim of the Work: Study the role of hepatic stellate cells and angiogenesis in liver regeneration after 70% partial hepatectomy in adult male albino rats.Materials and Methods: Forty adult male albino rats(4-6months) weighing 150-200 gm were randomized into 5 groups of eight rats each; group I was the sham control group, and the animals were subjected to 70% partial hepatectomy in groups II, III, IV and V in which they were postoperatively sacrificed 3, 7, 14 and 21 days respectively. At the time of scarification specimens were taken for histological and immunohistochemical studies. H and E and immunohistochemical staining for proliferating cell nuclear antigen (PCNA), vascular endothelial growth factor (VEGF) and α-smooth muscle actin (α- SMA) were done. Image analysis and statistical analysis of the obtained results were performed.Results: The liver showed disturbed histological architecture with distortion of the neatly arranged hepatocyte plates in group II which was restored in the following groups. Wide distribution of Kupffer cells, was also noted in group II and continued in group III. The PCNA and VEGF activity showed significant positive reaction in groups II and III , insignificant positive immunoreaction was reported in groups IV and V . The α- SMA showed significant positive reaction in group II, insignificant immunoreaction was reported in the remaining other groups.Conclusion: Partial hepatectomy stimulated liver regeneration in the remaining liver lobes with reconstitution of most of the liver mass 21 days postoperatively. Abstract

简介:肝部分切除术是外伤性肝实质破裂和肝细胞癌的主要治疗方法。肝星状细胞(HSCs)在肝脏再生中发挥着重要的作用。工作目的:研究成年雄性白化大鼠70%肝部分切除后肝星状细胞和血管生成在肝再生中的作用。材料与方法:40只体重150 ~ 200 gm的成年雄性白化大鼠(4 ~ 6月龄)随机分为5组,每组8只;I组为假对照组,II、III、IV、V组取70%肝部分切除,分别于术后3、7、14、21 d处死。在划伤时,取标本进行组织学和免疫组织化学研究。进行增殖细胞核抗原(PCNA)、血管内皮生长因子(VEGF)和α-平滑肌肌动蛋白(α- SMA)的H、E和免疫组化染色。对所得结果进行图像分析和统计分析。结果:ⅱ组肝组织结构紊乱,肝细胞板排列整齐,各实验组均恢复原状。Kupffer细胞广泛分布于II组,并在III组继续存在。PCNA和VEGF活性在II组和III组呈显著阳性反应,IV组和V组免疫反应不显著。α- SMA在ⅱ组呈显著阳性反应,其余各组均无明显免疫反应。结论:肝部分切除术可促进剩余肝叶的肝脏再生,术后21天大部分肝肿块得以重建。这一再生过程与血管生成和造血干细胞激活相关,分别由VEGF表达和α- SMA表达表示。
