Investigation of the effect of mulberry leaves powder concentration (1~5%), using the DNJ high-production bacteria, provided evidence for microbial mass production of DNJ. When the 4% mulberry leaf powder for 9 days was used, the α -glucosidase inhibitory activity was over the 85%. Abstract

1-Deoxynojirimycin (DNJ) have been extensively investigated for their α -glucosidase inhibitor on postprandial hyperglycemia, and applied in nutraceuticals and medicine for preventing or delaying progression of type 2 diabetes. α-葡萄糖苷酶抑制枯草芽孢杆菌培养条件的优化
1-脱氧诺吉霉素(DNJ)作为α -葡萄糖苷酶抑制剂对餐后高血糖的抑制作用已被广泛研究,并应用于预防或延缓2型糖尿病进展的营养保健和药物中。但桑叶中DNJ含量较低(约0.1%),需要更有效的提取方法。本研究旨在开发微生物DNJ,作为生物方法DNJ的替代方法。在本研究中,我们通过高效液相色谱法获得了枯草芽孢杆菌大量产生DNJ的证据。测定α -葡萄糖苷酶活性对DNJ产生或不产生的抑制作用。研究桑叶粉浓度(1~5%)对DNJ高产菌的影响,为微生物量产DNJ提供依据。4%桑叶粉处理9 d后,其α -葡萄糖苷酶抑制活性达到85%以上。此外,本研究的结果证实了微生物在使用各种营养物质时DNJ产量的增加,并为提高微生物DNJ产量的方法提供了见解。
