In this study, we characterize a simple cholesterol-short-interfering RNA (siRNA) conjugate system targeting <i>Tnf</i> mRNA in liver macrophages for the treatment of NAFLD. First, we observed delivery and anti-inflammatory activity in an acute liver inflammation model. In a follow-up murine NAFLD model, we observed total prevention of nearly all hallmarks of this disease: steatosis, inflammation, and fibrosis.
