Nemertea

Isolate a small 1-2cm long piece of the adult female (worms with brown coloration) using small scissors. In a dry petri dish make several lateral cuts in the body wall to release the eggs. Collect these using a pipette and transfer them into a small 35mm petri dish containing filtered sea water (FSW). Wash the eggs three times with FSW. Eggs can be concentrated by swirling them towards the center of the dish prior to transferring them to a new dish with fresh FSW. When first collected the eggs will contain a prominent germinal vesicle. Germinal vesicle breakdown (GVBD) takes approximately 30 mins. after which the eggs can be fertilized.
