The following process is to produce crude enzyme from Aspergillus niger and Trichoderma viride. The resulting crude enzyme cellulase activity of 0.774 U/mL. Then, the crude enzyme is immobilized by Chitosan-GDA. OPEFB hydrolysis process with immobilized cellulase was carried out for 5, 7 and 9 days at 37°C. 摘要

油棕空果束(OPEFB)是木质纤维素,由13.20-25.31%的木质素,42.70-65.00%的纤维素和17.10-33.50%的半纤维素组成。通过有机溶剂预处理和固定化酶水解相结合的方法,纤维素可以作为一种新的可再生能源,用于还原糖。采用有机溶剂预处理木质素,以环境友好、易于回收的乙醇为溶剂,浓度为S/L 10%(w/w),在160℃下发酵90-150 min,以黑曲霉和绿色木霉为原料制备粗酶。所得粗酶纤维素酶活性为0.774 U/mL。然后用壳聚糖- gda固定化粗酶。在37℃条件下,固定化纤维素酶对OPEFB进行5、7、9天的水解。在160℃、150 min条件下,木质素降解效果最好,木质素去除率达到56.68%,而纤维素水解率最高,在9天的处理时间内达到47.59%。
