摘要

全氟烷基物质全氟辛酸(PFOA)影响雄激素代谢的一种新的内分泌干扰特性已经被发现。这一发现对我们理解PFOA对雄激素作用的影响具有启示意义。在荧光光谱抑制剂筛选和抑制研究中,PFOA被鉴定为有效的、竞争性的和特异性的抑制醛酮还原酶1C2 (AKR1C2),该酶催化5α-二氢睾酮(5α-DHT)在纳摩尔范围内还原为无活性的类固醇3α-雄甾二醇。这种特性被发现是缬氨酸存在于装订袋中的结果。因此,PFOA抑制了5α-DHT的还原,导致其积累。pfoa通过抑制AKR1C2而阻断5α-DHT代谢,增加雄激素受体的转激活。
