Ugarova","doi":"10.1134/S0006350924700556","DOIUrl":null,"url":null,"abstract":"<div><p>The possibilities of using a test system based on live <i>E. coli</i> BL-21 (DE3) Codon Plus cells expressing pH-resistant <i>Luciola mingrelica</i> firefly luciferase to study the kinetics of the effect of aminoglycosides (using the example of gentamicin) on the content of ATP and luciferase inside and outside the cells were demonstrated. It has been shown that after 3 h of incubation of bacteria with an antibiotic it was possible to assess changes in cell viability and the effectiveness of the antibiotic, as well as to predict the formation of persisters. 使用基于表达耐 pH Luciola mingrelica 萤火虫荧光素酶的活大肠杆菌 BL-21 (DE3) Codon Plus 细胞的测试系统,研究氨基糖苷类药物(以庆大霉素为例)对细胞内外 ATP 和荧光素酶含量的影响动力学的可能性。结果表明,在细菌与抗生素培养 3 小时后,就可以评估细胞活力的变化和抗生素的有效性,并预测宿主的形成。该方法有望用于对抗菌剂和药物形式进行快速、高效的初级筛选,以评估其有效性和作用机制。
