The expression of V6 was confirmed by analyzing the promoter activity of V6 ORF upstream sequences and quantifying the viral DNA accumulation in V6-mutant MCLV-infected tomato plants. Infectious clones of V6-mutant MCLV vII (pCA-1.1MCLV<sup>mv6</sup>) and V4 and V6 double-mutant MCLV vII (pCA-1.1MCLV<sup>mV4mV6</sup>) were constructed, and experiments transfecting protoplasts isolated from <em>Nicotiana benthamiana</em> and infecting tomato using the constructs were performed. The results indicate that V6 plays a critical role in the multiplication of MCLV in protoplasts, although it has minimal, if any, impact on the systemic infection of MCLV in plants.
