摘要

循环肿瘤细胞(Circulating tumor cells, CTC)计数在临床上已被作为判断不同肿瘤预后的重要指标,但目前许多CTC的分离或鉴定方法尚无临床资料。在这项研究中,我们建立了一种新的针对EpCAM或细胞角蛋白的间接免疫荧光染色方法来鉴定CTC。选取20例健康献血者和36例肿瘤患者的外周血标本进行截止建立和敏感性试验。EpCAM或细胞角蛋白染色的免疫荧光强度阈值为5080,可获得高度准确的细胞分类(灵敏度95%,特异性>99%),以2个CTC为截断点,CTC检测对不同癌症患者的诊断灵敏度为54.2%(19/35)。此外,ctc的存在(定义为bbb2.0 ctc /7.5 mL)可能与癌症分期有关,并且在乳腺癌患者中出现的时间比癌症复发早3周,因此被认为是监测化疗期间肿瘤复发的临床有用工具。
