The blood glucose concentration significantly changed through time for all three doses (P<0.0001). No significant differences in protein concentration and cell count in synovial fluid were found between treated and control joints (P>0.05), no significant difference in synovial glucose concentrations was found between treated and control joints (P>0.05). Values obtained by HPLC analyzed with PkAnalyst program revealed that the pharmacokinetic values were dose dependent, there was no significant difference in concentration of blood glucose between the three different doses (P>0.05). ELISA for IGF-1 (insulin-like growth factor 1) revealed a significant difference between treated and control joints (P<0.001). 摘要

胰岛素诱导马软骨细胞有丝分裂并促进II型胶原的生成。当关节内注射胰岛素时,会改变滑液的成分,包括葡萄糖浓度、胰岛素和血糖水平。采用高效液相色谱法测定关节内胰岛素浓度,得到药动学值。6匹杂交马分别在腕前关节处注射3种不同剂量的胰岛素(10、15和20 IU),对侧关节处注射等渗盐水。三种剂量组小鼠的血糖浓度随时间变化均有显著性差异(P < 0.05),治疗组与对照组关节间滑膜葡萄糖浓度无显著性差异(P < 0.05)。采用PkAnalyst程序进行高效液相色谱分析,药代动力学值呈剂量依赖性,三种不同剂量间血糖浓度无显著差异(P < 0.05)。胰岛素样生长因子1 (IGF-1) ELISA检测结果显示,治疗组和对照组关节间存在显著差异(P<0.001)。本研究中使用的胰岛素对马的关节是无害的,对于350-400公斤的马,不应施用超过20 IU。
