QRT-RCR was used to assess the expression of bax , bcl2, casp3, and casp9 genes, and flow cytometry was used to evaluate apoptosis in cancer cells. Results: The inhibitory effect of dose- and time-dependent B. coagulans supernatant showed that the supernatant of this probiotic bacterium had a cytotoxic effect on SKBR3 cancer cells. On the other hand, analysis of flow cytometry results and increased expression of bax , casp3, and casp9 pro-apoptotic genes and decreased bcl2 expression in cancer cells showed induction of apoptosis. 摘要

背景与目的:癌症是伊朗妇女最常见的癌症类型之一。到目前为止,通常的癌症治疗还没有完全有效。因此,开发抗癌产品具有重要意义。本研究的目的是评估凝结芽孢杆菌益生菌上清液对SKBR3细胞的细胞毒性、抗癌和诱导作用。材料和方法:采用MTT法对不同浓度的益生菌上清液(1、2、3、4、5、6和7mg/mL)在SKBR3细胞上24、48和72小时的抗癌潜力和细胞毒性作用进行评估。采用QRT-RCR检测bax、bcl2、casp3和casp9基因的表达,流式细胞术检测癌症细胞凋亡。结果:混凝芽孢杆菌培养上清对SKBR3癌症细胞具有一定的细胞毒性作用。另一方面,流式细胞术结果的分析和癌症细胞中bax、casp3和casp9促凋亡基因表达的增加以及bcl2表达的降低显示了细胞凋亡的诱导。结论:凝结芽孢杆菌益生菌上清液对SKBR3癌症细胞的抗癌和细胞毒性作用表明,随着研究的深入,该益生菌可作为治疗癌症的新策略。
