摘要

目的采用高效液相色谱法测定不同产地淫羊藿中7种主要黄酮类化合物的含量。方法高效液相色谱条件为:SHISEIDO mg - c18色谱柱(3.0mm×100 mm,3.0μm),流动相为乙腈-含0.1%甲酸的水为梯度洗脱液,乙腈梯度为25%(0-10min)、25%-40%(10-12min)、40%-45%(12-22min)、45%-75%(22-25min)、75%(25-30 min),流速为0.6 mL/min,紫外检测波长为270 nm,柱温为25℃,进样量为5μL。用70%乙醇溶液超声提取样品。结果淫羊藿苷A、淫羊藿苷B、淫羊藿苷C、淫羊藿苷、淫羊藿苷Ⅰ、淫羊藿苷Ⅱ、无氢淫羊藿苷7种黄酮类化合物均在30 min内分离得到,标准曲线线性良好(r=0.999 9),日内、日间精密度RSD%均小于2.0%,回收率为98% ~ 102%,稳定性和重复性RSD%均小于2.0%。结果表明,该方法能够满足方法验证的要求。对辽宁、甘肃、湖北三地淫羊藿药材和对照药材中7种黄酮类化合物的含量进行了测定。结论本方法快速、简便,可用于淫羊藿药材的质量控制,为淫羊藿总黄酮的药动学及组织分布研究奠定基础。
