The results of DSC and XRD studies revealed the molecular amorphous dispersion of Sildenafil citrate into the gellan gum microspheres. Microspheres so prepared were discrete, bulky, free flowing and showed an average encapsulation efficiency ranging from 95-98%. The formulation exhibited a good mucoadhesive strength which was determined in in vitro conditions through falling film technique and was compared with ex vivo studies. The microspheres so prepared also exhibited a good swelling index which confirmed the strong mucoadhesive property of the formulation. 摘要

本研究的目的是制备枸橼酸西地那非的结冷胶微球,用于鼻内给药,以避免第一次代谢。采用喷雾干燥法制备微球。对微球的粒径、掺入效率、溶胀能力、zeta电位、体外黏附、体外黏附、热分析、XRD研究和体外药物释放等特性进行评价。对不同动力学方程的体外数据处理表明,结冷胶微球的药物传递受扩散控制。DSC和XRD研究结果表明,枸橼酸西地那非在结冷胶微球中的分子无定形分散。制备的微球离散、体积大、流动自由,平均包封效率在95-98%之间。在体外条件下,通过降膜技术测定了该制剂具有良好的粘接强度,并与离体研究进行了比较。所制备的微球还表现出良好的膨胀指数,证实了该配方具有较强的粘接性能。关键词:结冷胶,微孢子,喷雾干燥,柠檬酸西地那非,鼻给药
