Group II, III, and IV rats were orally administered the PHF at dose level of 26.25, 52.5, and 105 mg/kg once a day, for 15 days, respectively. Group V was orally administered 2.7 mg domperidone. On 16th day, rats were sacrificed. Mammary glands were isolated and processed for mRNA expression analysis using real-time polymerase chain reaction (qRT-PCR). The results demonstrated that the mRNA expression of ACTA2 and CK14 increased in dose-dependent manner in the groups of PHF. Significantly different between the Group III, IV, and V compared to Group I was observed (p 0.05). 摘要

在动物实验中,含有雌雄同体索罗、三角Trigonella foenum-graceum和辣木提取物的复方中药制剂(PHF)具有一定的促乳作用。然而,其分子作用尚未被阐明。本实验旨在探讨PHF对泌乳大鼠乳腺肌上皮细胞α-肌动蛋白平滑肌(ACTA2)和细胞角蛋白14 (CK14) mRNA表达的影响。30只雌性Wistar大鼠分为5组,每组6只。第一组口服aquadest。II、III、IV组大鼠分别以26.25、52.5、105 mg/kg的剂量口服PHF,每天1次,连用15 d。V组患者口服多潘立酮2.7 mg。第16天处死大鼠。分离乳腺,采用实时聚合酶链反应(qRT-PCR)进行mRNA表达分析。结果表明,PHF组ACTA2和CK14 mRNA表达呈剂量依赖性增加。III、IV、V组与I组比较差异有统计学意义(p < 0.05)。综上所述,PHF增加了泌乳大鼠乳腺肌上皮细胞ACTA2和CK14 mRNA的表达。
