Abstract
A new and improved procedure has been developed for the isolation of intact DNA genomes from purified vaccinia virions. Purified virions are layered on a neutral sucrose gradient containing sodium dodecyl sulfate, 2-mercaptoethanol and sodium chloride at neutral pH. Intact viral DNA free from protein and fully sensitive to DNase I is rapidly released from the virions.
