摘要DNA元条形码已成为生物多样性评价和监测的常用工具。DNA元条形码的结果如何与基于“经典”的研究进行比较,在大多数情况下形态学物种鉴定,仍然不清楚。根据已知基线调查物种检测的研究实际上是不存在的。在这项研究中,我们利用在奥地利东部采集的光诱器样本,利用Illumina MiSeq平台调查了形态学物种列表与COI元条形码结果之间的一致性。比较了两种不同长度的引物组合(313 bp和205 bp)对扩增子长度的影响。物种检出率在0.38 ~ 0.69之间;与较长的扩增子相比,较短的扩增子平均具有较高的物种检出率。通过元条形码检测到的单一种较少。一个物种被发现的主要决定因素是它的生物量,即较小的物种被发现的机会较低。然而,也有证据表明在超家族水平上存在分类偏差。虽然生物量的影响是可以预料的,但分类学偏差的存在引起了关注,需要进一步的研究。当元条形码用于确定保护措施时,这种偏差可能是重要的。对所有样点的排序分析表明,就群落生态学而言,我们的元条形码结果基本保留了整个物种列表所获得的总体格局。
