Abstract

Acute liver failure (ALF) often progresses rapidly and ultimately causes a high mortality due to the lack of timely and effective treatment strategies. Transplantation based on adipose-derived mesenchymal stem cells (ADSCs) has shown strong potential in treating ALF. However, the efficacy of ADSCs transplanted into vivo is often unsatisfactory, which seriously affects the therapeutic effect on ALF. Many studies have proven that tFNAs, as novel nanomaterials, regulate the function of cells. In this study, we evaluated the functional changes in ADSC proliferation, migration and hepatogenic differentiation after treatment with tFNAs. Relevant results demonstrated that tFNAs improved ADSCs proliferation and migration. In addition, combined with biological informatics, we comformed that tFNAs can improve hepatogenic differentiation of ADSCs via activating the MAPK signalling pathway. Furthermore, in vivo experimental studies with ALF model mice showed that tFNAs not only increased the number of ADSCs migrating to the damaged liver but also significantly improved the effectiveness of ADSCs in the repair of liver tissue. In conclusion, tFNAs combined with ADSCs were used in the treatment of ALF for the first time, which defines a promising method to improve the efficiency of ADSC transplantation in the treatment of ALF.
