Untuk memenuhi permintaan pasar maka stok bibit harus terpenuhi salah satunya dengan menggunakan metode vegetatif. Nutrien merupakan salah satu faktor penting dalam proses metode ini. Jika media tumbuh tidak mencukupi kebutuhan perlu dilakukan pemupukan. Penelitian ini bertujuan untuk mengetahui pengaruh variasi dosis media Walne pada pertumbuhan Gracilaria sp. dan mengetahui dosis optimal sebagai sumber nutrien. Metode yang digunakan adalah metode eksperimental laboratoris dengan Rancangan Acak Lengkap (RAL) menggunakan 5 perlakuan 1 kontrol dan 3 ulangan, antara lain perlakuan A (kontrol), B (0,5 ml/L), C (1 ml/L), D (2 ml/L), E (4 ml/L) dan F (8 ml/L). Parameter yang diamati adalah berat eksplan, jumlah tunas, panjang tunas, pertumbuhan berat mutlak, laju pertumbuhan spesifik dan kualitas air media. Data dianalisis menggunakan analisis One-Way ANOVA dilanjutkan uji Tukey dan Games-Howell. Hasil penelitian menunjukkan rata-rata berat, jumlah tunas dan panjang tunas terdapat perbedaan yang signifikan (p < 0,05). Rata-rata pertumbuhan berat mutlak tertinggi pada perlakuan E (4 ml/L) yaitu 0,07 ± 0,01 g/hari dan laju pertumbuhan spesifik tertinggi pada perlakuan E (4 ml/L) 2,24 %/hari. Hal ini diduga nutrien bekerja secara optimal. Hasil pengamatan pH (4 – 7,92), salinitas (23 – 32 ppt) dan suhu (27 – 31 oC). Dosis 4 ml/L merupakan konsentrasi optimal karena pada dosis ini Gracilaria sp. berpengaruh secara signifikan. Gacilaria sp. is a source of agar which is very salable in the market both domestically and abroad. To meet market demand, the stock of seeds must be met, one of them by using the vegetative method. Nutrients are one of the important factors in the process of this method. If the growing media is not sufficient, fertilization needs to be done. This study aims to determine the effect of variations in the dose of Walne media on the growth of Gracilaria sp. and knowing the optimal dose as a source of nutrients. The method used is a laboratory experimental method with Completely Randomized Design (CRD) using 5 treatments 1 control and 3 replications, including treatment A (control), B (0,5 ml/L), C (1 ml/L), D (2 ml/L), E (4 ml/L) and F (8 ml/L). Parameters observed were explant weight, number of shoots, shoot length, absolute weight growth, specific growth rate and water quality of the media. Data were analyzed using One-Way ANOVA analysis followed by Tukey and Games-Howell tests. The results showed that there were significant differences in the average weight, number of shoots and shoot length (p < 0,05). The highest average absolute weight growth was in treatment E (4 ml/L) which was 0,07 ± 0,01 g/day and the highest specific growth rate was in treatment E (4 ml/L) 2,24%/day. It is suspected that nutrients work optimally. The results were observed for pH (4 – 7,92), salinity (23 – 32 ppt) and temperature (27 – 31 oC).
