Morphological and biochemical evaluation of effects of Myrtus communis L. extract on heart and aorta in high fat-diet-induced obese rats

Triglyceride, cholesterol, and high-density lipoprotein (HDL) were estimated in blood serum. Tissue oxidative stress and\ninflammatory parameters were evaluated biochemically. Tissue morphologies, eNOS, inducible NOS (iNOS), and NADPH oxidase-2\n(NOX-2)-immunopositive and apoptotic cells were evaluated histologically.\nResults: Altered serum lipid profiles, degenerated heart, and aorta morphology, increased malondialdehyde, 8‐hydroxy‐2‐\ndeoxyguanosine, tumor necrosis factor-alpha, monocyte chemoattractant protein-1 and myeloperoxidase levels, and iNOS, NOX-2\nimmunopositive and apoptotic cells, decreased NO levels, eNOS-immunopositive cells in both tissues were observed in HFD group.\nAll these parameters improved in the HFD+MC group.\nConclusion: This study revealed that HFD-induced obesity increased iNOS activation and oxidative stress in the cardiac and aortic\ntissues of the rats. Abstract

Objective: The purpose of this study was to examine the protective effects of Myrtus communis L. (MC) extract on high fat-diet (HFD) induced heart and aorta damage by evaluating oxidative stress and the endothelial nitric oxide system (eNOS). Materials and Methods: Wistar albino male rats were divided into 3 groups (n=7) as control, HFD, and HFD+MC. 桃金娘提取物对高脂饮食致肥胖大鼠心脏和主动脉影响的形态学和生化评价
目的:通过评价氧化应激和内皮型一氧化氮系统(eNOS),探讨桃金子提取物(MC)对高脂饮食(HFD)诱导的心脏和主动脉损伤的保护作用。材料与方法:Wistar雄性白化大鼠分为3组(n=7),分别为对照组、HFD组和HFD+MC组。HFD组和HFD+MC组大鼠以HFD喂养16周,后4周口服生理盐水或mc100 mg/kg(5天/周)。测定血清中甘油三酯、胆固醇和高密度脂蛋白(HDL)。用生化方法评价组织氧化应激和炎症参数。组织形态学、eNOS、诱导NOS (iNOS)、NADPH氧化酶-2(NOX-2)免疫阳性细胞和凋亡细胞进行组织学评价。结果:HFD组血清脂质谱改变,心脏和主动脉形态变性,丙二醛、8 -羟基-2 -脱氧鸟苷、肿瘤坏死因子- α、单核细胞化学引诱蛋白-1和髓过氧化物酶水平升高,两组组织中iNOS、nox -2免疫阳性和凋亡细胞,NO水平降低,enos免疫阳性细胞。HFD+MC组上述指标均有改善。结论:本研究揭示了hfd诱导的肥胖增加了大鼠心脏和主动脉组织中iNOS的激活和氧化应激。MC改善了氧化/抗氧化平衡,并通过eNOS参与防止心脏和主动脉损伤。
