肉仔鸡肝脏热应激反应的CUT&tag-seq分析为日粮根皮素提高耐热性提供了新的见解
本工作旨在通过靶下切割和标记结合高通量测序(CUT&;Tag-seq),研究根皮素通过核因子-红系2相关2(Nrf2)-DNA相互作用对热应激肉鸡肝脏抗氧化特性的影响。将160只22日龄Arbor Acres肉鸡分为4组:常温组(NT:22.8–23.4°C,湿度46–55.8%)饲喂基础日粮,100mg/kg(LT组)和200mg/kg(PT组)根皮素。取42日龄肉鸡的肝脏样品。高温条件下丙二醛(MDA)水平升高(P≤0.05),谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和Nrf2的活性和mRNA表达水平降低(P≤0.05。日粮200 mg/kg根皮素可降低(P≤0.05)热应激肉鸡肝脏MDA水平,但可提高(P≤0.01)GSH-Px、SOD、CAT和Nrf2的活性和mRNA表达水平。CUT&;Tag-seq显示,与HT组相比,PT组有4042个DEP上调,而2322个DEPs下调。DEPs的Nrf2结合基因包含SOD3、CAT、SOD2和p38丝裂原活化蛋白激酶。大多数TOP20重要的KEGG途径与“环境信息处理”有关。总之,根皮素可以改善热应激肉鸡肝脏中Nrf2及其下游DNA位点(包括基因和Nrf2结合的非编码RNA)的氧化损伤。
