Abstract

Dans cet article, on a cherché à préciser les facteurs qui affectent la qualité et à établir les critères de validation du dosage immunologique.

La spécificité, ou l'aptitude à pouvoir discriminer des substances structurellement différentes de l'analyte, dépend du choix du site de couplage à la protéine immunogène et de la diversité antigénique du milieu biologique. L'introduction par exemple d'un bras de couplage engendre dans certains cas l'existence de populations d'anticorps anti-bras qui restreignent à la fois la spécificité et la sensibilité.

La senslbilité théorique d'un système est limitée par l'affinité des anticorps et l'activité spécifique du traceur. Dans un système compétitif, celle-ci peut ētre définie par la pente de la courbe dose-réponse et/ou la dose produisant un signal différent de celui obtenu en l'absence de compétiteur.

Des exemples illustrent les problēmes rencontrés dans le cas de la testostérone, de la digoxine et de la méthlonineenképhaline.

Mit diesem Artikel wird bezweckt, diejenigen Faktoren, welche der Qualität immunologischer Bestimmungen abträglich sind, näher zu erörtern, und Bewertungskriterien festzulegen.

Spezifität, d.h. die Fähigkeit, Substanzen zu unterscheiden, die vom Analysanden strukturmässig verschieden sind, hängt von der Auswahl des Kopplungsorts am immunogenen Protein und der Antigenvielfalt des biologischen Milieus ab ; zum Beispiel löst die Einführung eines Kopplungszweigs in bestimmten Fällen das Vorhandensein entsprechender Antikörperpopulationen aus, die sowohl die Spezifität als auch die Empfindlichkeit einschränken.

Die theoretische Spezifität eines Systems wird durch die Affinität der Antikörper und der spezifischen Aktivität des Tracers begrenzt. In einem kompetitiven System kann sie durch eine Steilheit der Dosis-Reaktions-Kurve und/oder durch diejenige Dosis bestimmt werden, die ein anderes Signal erzeugt, und dies bei Abwesenheit interferieren der Substanzen.

Die bei Testosteron, Digoxin und Methionin-Enkephalin auftretenden Probleme werden anhand von Beispielen erläutert.
