摘要

近年来,抗生素耐药性以惊人的速度蔓延,对全球公共卫生构成严重威胁,导致全世界数百万人死亡。耐多药(MDR)微生物的出现是由于耐药的广泛传播和不同种类的细菌之间共享耐药基因。一种治疗难以治疗的细菌感染的有希望的策略是开发基于纳米材料的疗法,这种疗法可以绕过与获得耐药性相关的现有途径。本研究的目的是制备壳聚糖/果胶包封的紫锥菊提取物,并评价其对MDR菌株的抑菌效果。本研究采用凝胶化技术将苍鹭叶提取物包封成壳聚糖/果胶纳米颗粒(NPs)。利用扫描电子显微镜(SEM)、动态光散射(DLS)、透射电子显微镜(TEM)和傅里叶变换红外光谱(FT-IR)对合成的NPs进行了分析。研究了纳米颗粒对金黄色葡萄球菌的抗菌和抗生物膜活性。此外,还评价了合成NPs对hek93细胞的毒性。FT-IR光谱证实了官能团之间的相互作用。CS/PN NPs为球形,表面均匀,尺寸在80 ~ 110 nm之间。白芷提取物的PDI为0.521,包封率(EE%)为84.35%。合成的CS/PN NPs对公共卫生相关细菌具有抗菌和抗生物膜活性。此外,结果表明,含NPs提取物对HEK-93细胞无毒性影响。本文提出的研究结果将有助于开发具有更高稳定性和抗菌活性的新型植物提取物,从而减少对抗生素的需求。
