The posttransplantation diabetes mellitus(PTDM) is a frequent complication in immunosuppressive therapy. The present study was to examine the effect of a crude water extracts of medicinal herbs such as Sanguisorba officinalis(SOE) on the immunosuppressive activity with lymphocyte and insulin secretion in insulinoma cell lines with RIN-5mF. It was found that SOE treatment had effect of immunosuppressor on lymphocytes and also significantly increased insulin secretion in RIN-5mF compared to other agents. we might suggest a mechanism on insulin secretion by HNF4a. 摘要

免疫抑制剂环孢素A(CsA)和他克莫司(FK506),其主要细胞靶点是钙调神经磷酸酶/活化T细胞核因子(NFAT)信号通路,可降低β细胞胰岛素含量和mRNA表达。移植后糖尿病(PTDM)是免疫抑制治疗的常见并发症。本研究探讨了三地血(SOE)等中药粗提物对含RIN-5mF的胰岛素瘤细胞株淋巴细胞免疫抑制活性和胰岛素分泌的影响。结果发现SOE治疗对淋巴细胞具有免疫抑制作用,且与其他药物相比,显著增加了RIN-5mF的胰岛素分泌。我们可能会提出HNF4a影响胰岛素分泌的机制。综上所述,本研究提示SOE可能作为PTDM的免疫抑制药物。关键词:地榆,免疫抑制剂,胰岛素分泌,HNF-4α
