The hCOLIII at concentrations of 0.5, 0.25, and 0.125 mg/mL all showed specific effects on the proliferation and migration of human vascular endothelial cells. It could also affect the cell cycle, increase the proliferation index, and increase the expression level of VEGF in human vascular endothelial cells. In the meantime, hCOLIII at the concentration of 0.5 mg/mL also showed a promoting effect on vessel formation. hCOLIII can potentially promote the endothelization process of blood vessels, mainly by affecting the proliferation, migration, and vascular-like structure of human endothelial cells. At the same time, hCOLIII can promote the expression of VEGF. 摘要

方法:MTT法检测hCOLII对EA.hy926细胞增殖的影响,划痕法检测hCOLII对细胞迁移的影响,流式细胞术检测hCOLII对细胞周期和凋亡的影响,血管生成法检测hCOLII诱导EA.hy926细胞血管生成的能力,RT-PCR法检测hCOLII对VEGF表达的影响。结果:0.5mg/ml、0.25 mg/ml和0.125 mg/ml浓度的hCOLIII对人血管内皮细胞的增殖和迁移均有一定的影响。影响人血管内皮细胞的细胞周期,增加PI指数,提高VEGF的表达水平。同时,0.5mg/ml浓度的hCOLIII对血管形成也有促进作用。结论:hCOLIII具有促进血管内皮化的潜力,主要通过影响人内皮细胞的增殖、迁移和血管样结构。同时,hCOLIII能促进VEGF的表达。这种胶原蛋白证明了它作为心血管植入物原料的潜力。
