摘要

本研究试图在山西省范围内选择具有产油潜力的候选微藻。对32株微藻进行了检测。其中29株微藻来自运城丰河和盐湖地区4个地点的水样。从水生生物研究所淡水藻类培养馆藏中购买了3株微藻。采用微采摘法和铺板法对山西产微藻进行分离纯化。32株微藻的生物量在48.9 ~ 422.2 mg/L之间,以干重测定。氯仿-甲醇萃取法是传统的总脂质测定方法,尼罗红荧光法是测定中性脂质的新技术。本研究采用氯仿-甲醇萃取法和尼罗红荧光染色法联合测定32株微藻的脂质含量。结果表明,所有菌株的总脂质含量在5.4% DW ~ 30.1% DW之间,单位体积荧光值在4.1 ~ 181.5之间。高脂质产率是生物柴油品种选择的关键标准。根据微藻的产脂能力,我们筛选出了3株可能在山西有产油潜力的微藻。这3株菌株分别为NY017(盐杜氏菌)、NY023(线性尼氏菌)和NY025(古尼氏菌)。脂质产量分别为3.25 mg·L-1·d-1、3.03 mg·L-1·d-1和2.11 mg·L-1·d-1。
